As a former feis mom from the southeast who now lives in CO, I'll just add that it depends on the person. We've had very fit teens visit who tried their usual run the day after arriving and felt like they hit a wall. We've had a 40-year-old go right from the plane to hiking Red Rocks (stupid) and got very sick that night. One twenty-something got sick on a hike on Day-2 and a second one felt nothing. And we've had 60-year-olds visit who never felt any ill effects. Even those who had problems with the altitude felt fine by Day 3, and those problems were mostly headaches. As others said, drink lots. Start hydrating before leaving and keep it up. And if you can, arrive a couple of days early to acclimate. Adults - skip the alcohol for a few days.
