Not without context. My patience had just reached a boiling point after the cynical start of what would have become another unnecessary debate between adherents of Kashi and Shibendu.

Amen generally means, in the USA, that you agree and are ending the conversation.

No, used in a discussion it means that your opinion is irrelevant and the conversation is ended.
