The couplet at the end alters the meaning of the sonnet by telling how rare Shakespeare's mistress is, she is not what she appears to be. Basicaaly she turned out to be quite different than expected.

He mocks aspects of the traditional sonnet because you notice his last couplet, first of all, has the last words at the end of each line rhyme. Second of all, you also notice he is not using his iambic metameter like he is in the other lines of the sonnet. Iambic meter basically has a da-dum,da-dum kind of beat and Shakespeare usually uses it, but at the last couplet he didn't.
