HoD was where Frank finally revealed as fact that the axoltl tanks were Tleilaxu women.

In earlier books it was mentioned that Tleilaxu women had never been seen by outsiders. Other clues were dropped and allusions made. But on pg 420 of the Hardcover Mother Superior Taraza says to the Tleilax Master Waff "None of us is now nor will ever become..." she paused ".... an axlotl tank."

That was, I believe, the first time Frank specifically said what axlotl tanks were.

In the cases you mentioned, axlotl tanks are indeed women. C:D... well I'm not sure about I'm just starting to re-read that now. I don't remember if the Bene Gesserit succumbed to using axlotl tanks.

I'm assuming with the Prelude to Dune series Brian figured that if you read his Father's books you'd know this, and if you didn't he'd build up to it til revealed in House Corrino.
