It is pretty cryptic at this point. Closest I can guess is that the Twilight plan is to eliminate all the Slayers, and Buffy is their "First", the leader. I suspect that Roden (and those he is serving) assume that once Buffy is dead, it will be all that much easier to kill off the rest of the Slayers.

(It's pretty much a given that Gigi isn't in on this part of the plan, and I'd bet that Faith had it exactly right-- gather all the Slayers together to "bake into a bloody pie", Gigi included.)

One other more radical interpretation is that since Buffy was responsible for calling all these Slayers, her death would mean they'd all go back to being "normal" women, or would enable some kind of magicks to make that happen.

(Such as-- Buffy being the Slayer who pulled the Scythe from the stone, she is now mystically bound to it, and/or it to her. If she dies, it becomes possible for what would essentially be the reverse of Willow's spell, and the Slayer essences would be removed from the current carriers and put back into the Scythe. The Scythe could then be destroyed, and that would be the last of the Slayers.)

So it's the twilight of the gods? The "gods" being the Slayers?
