Well, when Buffy first staked the Ubervamp, she was probably applying the same amount of force she would use for a regular vampire, and so was shocked when the vamp didn't die, assumed stakes through the heart couldn't kill it and didn't try again. After Anya's little research digging, however, they knew all they had to do was stake them really, really hard.

It must also be considered that Buffy was pretty sleep deprived in her initial encounter with the Ubervamp, so it wasn't like she was fighting at her best. And let's not forget that the Turok-Han were fighting as an army in "Chosen", whereas in "Bring on the Night" Buffy fought a Turok-Han one-on-one. If action movies and comic books have taught us anything, it's that an anonymous bad guy will last longer and put up a much better fight on his own than as part of a large group. This was previously shown to apply to Buffy; note how in "Sleeper" and "Into the Woods" Buffy fights vampire packs and kills them in about the same amount of time it usually takes her to kill a single vampire when going one-on-one.
