以掀起系列作中一連串生化危機的大企業umbrella為主軸的原創新作，除了串聯各系列作已知的劇情之外，並將闡述umbrella最終如何崩潰的未知謎團。

遊戲的類型為槍枝射擊遊戲，將利用 Wii 遙控器的紅外線指標來進行如同傳統光線槍射擊遊戲的玩法，使用槍枝來消滅安布雷拉以生化技術所創造出的僵屍與各種異形生物，並可破壞場景中出現的各種物件，像是門、櫥櫃、吊燈...等等。之外還可以依照畫面的指示輸入指令，進行各種動作場面的演出。

遊戲登場角色橫跨各系列作，包括S.T.A.R.S隊員CHRIS，JILL，REBECCA，BILLY，以及擔任幕後黑手的S.T.A.R.S隊長WESKER等等。
