You could start with a beach around St. Pete or Treasure Island. Take the Sunshine Skyway Bridge over to I75 go south to Sarasota and stop at Siesta Key (Travel Channel has named one of the world's best beaches) and then head to Sanibel or Ft. Myers Beach. I love Sanibel but it's a very quiet area, I don't think of Sanibel as having a lot of nightlife. Maybe visit Sanibel during the day for beach and shopping but stay over near Ft. Myers Beach.
