Subject: SDS 14-200 kit (beckman coulter)

With the beckman CGE kit SDS 14-200 we often see a lot of spikes in time window of approximately 1-5 minutes. This happens always a few minutes after the migration time marker (Orange G).

Has somebody also seen this problem?

Beckman suggested these are air bubbles. And we can indeed reproduce a lot of spikes in the same time window by injecting air in the capillary.

According to beckman these spikes should originate from the heat generated in the capillary during the run. Lowering the temperature of the capillary and/or the voltage should reduce the heat generated during the run and so the formation of air bubbles. This experiment showed however no improvement.

Furthermore, when the bubbles are formed by heat I expect to see spikes during the whole run and not in a small time window. Injecting air in the capillary results in the exactly reproduces the spikes. So I think the air is introduced at the beginning of the run.

I have however no clue how this should happen. Does anybody have suggestions? Or has anybody seen this before?
