Subject: Need help with creating a dynamic comic world

I have a question about creating a dynamic comic world. I'm writing a script about a milquetoast who's life is turned upside down by an outrageous comic character. Using past screwball genre films as a model (ex: Cary Grant/Kathyrn Hepburn in "Bringing Up Baby," "What's Up Doc," etc.), I'm seeking to create a "sane world/insane character" dynamic (I hope I'm making this clear).

Okay, here's the question:

I'm setting the story in the world of television, specifically the behind the scenes of a hit TV show. The meek hero is the assistant to the pompous star. Another character enters the mix, who brings comic trouble to the meek character, but ultimately gives him the courage he needs to grow, etc.

If I set the story within a TV show, with a bunch of colorful eccentric characters, does this dull the comic opposition the main comic character brings? In other words, am I killing the comic conflict by creating an "insane world/insane character" dynamic. It's quite frustrating, because I love my characters, but I think I'm making everyone so "nutty" that it's killing the overall comic conflict. Anyway, I could use some outside opinions. I hope the question wasn't too convoluted. Thanks.
