I'd say quite a great deal, actually. For starters, the technology is completely different, in that it's far behind what is imagined in the other shows, which is what makes it different. This isn't a gleaming, shiny, new spaceship, but a gritty, not too pretty-looking cargo ship. And there's no warp drive, and not much in the way of weapons besides shotguns.

Also, the focus is on the characters alone, with the lack of aliens or space battles or the like, whereas all of the other shows you mentioned dealt more on an epic scale with either politics, wars, space exploration, meetings with creatures of different kinds and sorts, or all of these traits. Even the sci-fi with strong characters, like "Farscape" and "Babylon 5," is still not the same as this, where there are still epic-y concerns.

Another difference is the fact that the enemy is not a completely evil empire like in "Star Wars." The Alliance is not a good government, and is not always just...but neither are our heroes. They are bureaucratic, uncaring, and authoritarian, but that does not make it evil, but deeply flawed. These flaws are why the characters of "Firefly" do not support the government.

Yes, all the shows you mentioned and "Firefly" are space operas...but very, very different kinds.
