Hi Franco, I know this must seem awful daunting to you (especially if you don't know much about how the female cycle works). Let me try to tackle your questions in the order that you laid them out.
Yes, I have tried AI. I am a single woman who is desperate to have a child. I have tried AI (sperm received from a sperm bank)3 times and got pregnant 2 out of those 3 tries. Unfortunately though, I lost the pregnancies. I haven't begun to try again yet (just lost the last one last month)but I will soon. I don't think I will go through the sperm bank, though. They get to be expensive. So, I am going to try and find a "fresh" donor (crossing my fingers)!
There are some really good resources out there to help you understand how the female body works and how to time the inseminations. The book I have is by Barbara Kass-Annese and Hal C. Danzer and it's called "The Fertility Awareness Handbook:the natural guide to avoiding or achieveing pregnancy". I found it easy to read and very informative. Now because sperm does not live for very long once it is ejaculated (only days), the timing for insemination has to be pretty darn close because the female egg will live for only 24 hours once a woman ovulates. To have sperm ready and on demand to meet the timing factor, some people use a sperm bank to get their sperm (like me). Sperm banks, after the donor makes a donation, are able to freeze the little guys in a lab (unable at all to do at home in our freezers. Not cold enough and not quick enough) so the shelf life of lab sperm can reach years and still be viable when thawed (my samples had been donated in 1999 and I was able to get pregnant). Because you will be "delivering" fresh sperm to the couple, it's imperative that they get it and inseminate it as soon as possible. This means that you have time to give them only ONE (1) donation. You may be able to get a second and possibly a third to them. They would need to be about 12 hours apart b/c this is about how often it is recommended a woman inseminate herself during her fertile time: 2 to 3 times 24-36 hours before she ovulates.
Another very good resource for information is this very website. This site even has products to allow a donor to ship fresh sperm to a woman for insemination. Check it out.
My advice is to have your sister and her partner become knowledgable on your sister's partner's fertility cycle so that all they need to do is give you a heads-up on date of demand. All you need to do is fill a sample cup get that sample in the mail on time (if you're away).
