Fritz (Dwight Frye) is startled and drops the jar he has come to steal, then furtively grabs the other jar containing the criminal brain. He probably had no idea of the horrors that would be unleashed due to his error.

The first signs of life from the monster, followed by the almost maniacal cry of "It's Alive!" from Henry Frankenstein (Colin Clive).

The creation scenes in Frankenstein and Bride of Frankenstein, as Officer Torch mentioned. The Kenneth Strickfaden gadgetry enhances the atmosphere of tension and suspense.

There are many more great moments, including one from Bride of Frankenstein cited by Joystar in an earlier thread; the pained expression on the monster's face as he says "we belong dead" and pulls that lever.

I recently watched the Universal Frankenstein films and my favorite moment is still the monster's encounter with the blind hermit in "Bride of Frankenstein. Other moments In BRIDE would be The monster's encounter with Dr Pretorious in the crypt, the escape from the jail, and the climax when the bride rejects the monster. And I also liked Henry's reaction to Dr Pretorious's little people.

In the first Frankenstein film it would be the first appearance of the monster. Henry's "It's Alive." Henry trying to avoid the monster in the windmill.
The monster coming to as Dr Waldman is preparing to dissect him. The scene Tim mentioned with Fritz dropping the brain is also a good moment.

The best moments in "Son of of Frankenstein" are with Ygor, Wolf and Krogh. The best scene with the monster is when he encounters Wolf. And the monster's discovery of Ygor's (Presumed) dead body.
