James Whale's Frankenstein, along with Tod Browning's Dracula, introduced the sound horror film genre to the world. A few of many memorable moments include:

The stark opening graveyard scene.

The climatic laboratory sequence that ultimately gave life to the monster, with all the flickering gadgetry and equipment fully engaged.

The monster's finger moves.

Henry Frankenstein's ecstatic, almost manicical utterance, "It's alive!"

The monster's first steps, as Boris Karloff steps into movie immortality.

The shrill off-screen scream of Fritz, played by Dwight Frye.

The scene with Maria by the lake.

The final sequence.

Tim has pretty much covered everything.
I'll add:
The monster attacting Henry, and Dr Waldman injects the monster with a tranquilizer.

The monster comes to and strangles Dr Waldman while he is preparing to disect the monster.
