Anybody here please feel free to correct me if I'm wrong, but so far as I know, suppositories - which remain and melt in the rectum - are all well and good for stimulating peristalsis in the rectum and lower colon, but given that much constipation often originates higher up in the colon, if you really want to get "cleaned out" you would do well to take a "large" enema the night before your flight.

From what I've been told, the best way would be to start with a warm, soapy 2 quart enema, and then rinse with another clear water 2 quart enema, to which you've added a half teaspoon of salt.
