Personally, I'm a fan of the Masquerade and the Soap Bubble. The room for the Masquerade is fairly large, so as long as you can get in line somewhat early, you should be able to get in. Last year, we actually had chairs left over. The Soap Bubble is the Saturday night ACEN dance (there's another one on Friday - slower lounge style supposedly). Techno/trance/progressive is the music selection for the Soap Bubble, and last year was the biggest glowstick fest I had ever seen with probably about 90-95% of the floor lit up with sticks. I also like going to the industry panels to learn about what's coming out. This year, I'm also looking forward to a lot of the concerts on Friday.

Those are just my preferences. A lot of people go to meet certain guests (which there are plenty of really good ones), some people go mainly for the dealer's room, some for the vid rooms, some just go to hang out with friends, etc. It's all about what you want to see. The thing you may want to do is try to map out what you want to see beforehand. There are so many things going on, you'll have to pick and choose what you want to see. At the same time, try to be flexible since things always come up.

I made a plan my first year, which totally went out the window after I met some really cool people there and I sat and talked with them most of the time. Even with the change in plans, it was a total blast.
