Larry Berger's "Saturday Light Brigade" has a live audience from its studio in the Children's Museum almost every Saturday, except holidays. He didn't have one when he was at WYEP, though.

If you're talking about a seated audience, watching a program in an auditorium, that "went out with button shoes," to quote Stan Freberg.

I guess it might have been the Wilkens Amateur Hour, which began on radio and then was simulcast on WDTV. It lasted until 1958, according to this link:
