James Marsters certainly plays the scene with Riley as though he as an actor does not think Spike is the Doctor. Riley calling him "the doctor" elicits only confusion and annoyance. The mention of eggs in the basement elicits immediate guilt and concern. The actor clearly felt that spike saw those as unrelated querries, whereas to Riley they were the same querry.
